Job Description Summary
We are looking for a Senior Research
Investigator (SRI) under the direction of Dr. Ellen Pure, to assume the responsibility of decoding the cell-cell and cell-matrix
interactions in the tumor microenvironment of solid tumors including their impact on vulnerability to Immunotherapy, which is a rapidly
advancing modality for the treatment of many cancers. However, significant hurdles still remain, including complex multi-directional
heterotypic interactions between myriad populations in the tumor microenvironment, such as cancer-associated fibroblasts, tumor cells,
inflammatory and immune cells, cancer-associated endothelial cells and perciytes, as well as extracellular matrix that promote tumor
initiation, progression and metastasis.
This position is contingent upon funding.
Job Description
SRI will be
expected to carry translational studies of solid tumors, developing novel and utilizing state-of-the-art techniques to generate and test new
therapies and mechanisms of stromal-dependent tumorigenesis and therapeutic resistance. Requires >3 years experience in pre-clinical studies
of novel potential therapeutics (including multiple routes of drug delivery including intraveneous injection in mice) in xenograft,
syngeneic transplant and autochthonous tumor models as well as real-time 2-photon imaging of precision cut tumor slices. Skill in
quantification of tumor growth by ultrasound, bioluminescence image and caliper measurements is essential. Must have expertise in the
generation of human and mouse CAR T cells. Will collaborate with other Penn Investigators on the use of tumor-on-a-chip as orthoganol
approach to these technologies. Extensive experience in multi-parameter flow cytometry and multiplex immunofluorescence of tissue a use of
advanced digital imaging programs for quantitative analysis of the latter. Expertise in acquisition of second harmonic images and their
quantification is required. Generation and maintenance of novel compound mutant mouse colonies is required.
