Position Summary
The researcher will collect sperm cells from mouse, immunolabel them, and image the cells using superresolution tools. The goals of the research project are (1) to determine whether protein kinase A (PKA) localization during capacitation requires initial increase of cAMP or relies on post translational modifications caused by lysine acetylation; and (2) to analyze by dual color staining localization of AKAP4 and PKA, during the progress of capacitation.
The project is planned for a period of three months, from August to October 2023. We will work with CD-1 mouse spermatozoa. Specifically, we will utilize spermatozoa from the cauda region of the epididymis. The spermatozoa will be incubated in non-capacitating or capacitating media. In addition, media will be supplemented with PKA antagonists, PKA inhibitor sPKI, and PKA agonists. Additionally, we will study the effect anchoring inhibitors of PKA. The use of this pharmacological reagents aims to study how they modify PKA localization and function on the course of capacitation.
